IL—2、TNF—α和PGE_2对破骨细胞性骨吸收的作用

【摘要】 从新生兔的四肢长骨中分离出破骨细胞,将其与大小约6mm×6mm、厚约10μm的牛骨片共同培养。24小时后,培养液中分别加入IL-2(50U/ml,100U/ml);TNF-α(10 ̄(-10)M,5×10 ̄(-10)M,10 ̄(-9)M);PGE_2(100ng/ml);并做空白对照。培养40小时、64小6寸及1周,利用相差显微镜计数吸收陷窝数,通过图像分析系统计算吸收陷窝表面积。结果表明:IL-2能促进破骨细胞性骨吸收:TNF-α对分离破骨细胞性骨吸收无明显影响;PGE_2则对体外分离的破骨细胞具有抑制作用。

【Abstract】 AbstractOsteoclasts were mechanically isolated from one-day-old neonatal rabbit’s long bones and cultured onborine bone slices of approximately 6mm × 6mm in sizeand 1μm in thickness. after 24 hours,each bone slicewas placed in a well of 24-well culture plate containingIL-2(50u/ml,100u/ml), TNF-α(10 ̄(-10) M,5× 10 ̄(-10)M,10(-9)M),PGE2(100ng/ml ) in medium 199,orcontrol. When they had been cuItured for 40 hours,64 hours and 1 week, the number of resorptionlacunae on each slice was counted under phase-contrastminroscope.In the same time,pictures were takenand used to count surface area of lacunaeby In thesLame time pictures were taken and used to countsurface area of lacunae by Interreactive lmage Analysissvstem The initial results show:L-2 stimulatedosteoclastic bOne resorption; TNF-α has notsignificant effect on osteoclastic bone resorption; whilePGE2 inhibited osteoclastic bone resorption.
