【摘要】 利用体外消化结合不连续密度梯度离心的方法，由肿瘤组织内分离出肿瘤浸润性淋巴细胞（ＴＩＬ），并在重组人类白细胞介素２（ｒＩＬ－２）和／或重组肿瘤坏死因子α（ｒＴＮＦα）存在下进行体外培养。结果表明，低浓度ｒＴＮＦａ可以促进ＴＩＬ体外增殖、增加处于Ｓ期及Ｇ＿２＋Ｍ期的ＴＩＬ比例、增加细胞毒Ｔ细胞比例、上调ＴＩＬＩＬ－２Ｒ表达，并能明显增强ＴＩＬ体内外对自体及同种异体肿瘤的杀伤活性。更多还原

【Abstract】 umor-infiltrating lymphocytes(TILs ) were isolated from 8 solid tumors by means of in vitro di-gestion a nd d iscontinuous density grad ient centrifugation。 By incubating TILs in the presence of 6000Iu/ml rlL-2 and 500u/ml rTNFa,It’s demonstrated that rTFNa could enhance the proliferation ofTILs,promote the proportion of TILs in S-phase and G2 M-phase,enhance the percentage of CD3 +and CD8 + cells,up-regulate the expression of IL-2R,as well as enhance the cytotoxicity of TILs a-gainst autoIogous and allogenic tumors both in vitro and in vivo. The results are of significance for in-creasing immunotherapy effects and decreasing the adverse effects of certain cytokines.更多还原
