反相离子对高效液相色谱法分离测定川乌和附片中乌头类生物碱的方法研究

【摘要】 本文建立了反相离子对高效液相色谱法分离测定川乌、附片中3种重要生物碱。本法分离度好、灵敏度高、选择性好、准确、快速。

【Abstract】 RP-ion-pair HPLC was developed for the separation and quantitative determination of meaconitine,aconitine and hypaconitine in the Chinese medicine Chuan Wu and Fu Pian.The chromatographic conditions were:column: Shim-Pack ODS 150mm× 6mm ID,mobile phase:CH3OH-H2O-CH3CN(67:33:2.6m mol/L SDS,0. 02mol/L NaH2PO4,pH=4.5±0.2).detector:UV 235nm,flow rate:1.0ml/min,column temperature:35℃,Under the above conditions,three kinds of aconite alkloids were separated from each other as Well as from the interfering components in the sample.The suitability of chromatographic system was examined.The linear ranges for meaconitine,aconitine and hypaconitine were 0.0258~0.348μg(r=0.9997),0.0174~0.348μg(r=0.9996)and 0.023~0.46μg(r=0.9996)respectively.The method is simple,rapid.specific,highly sensitive,precise and reproducible.
