北五味子及其制剂三苓丹胶囊中五味子乙素的薄层光密度法测定

【摘要】 本文报道了用薄层光密度法测定北五味子及其制剂三苓丹胶囊中五味子乙素的含量。样品用乙醚回流提取2h,过中性氧化铝柱后用硅胶GF254板,以石油醚(30~60℃)-甲酸乙酯-甲酸(15:5:1)为展开剂进行薄层层析;λ=254nm、λ=400nm进行反射法锯齿扫描。本法灵敏度高,重现性好。

【Abstract】 A method of TLC-densitometry was developed to determine the active ingredient(γ-schisandrin)in Chinese magnoliavine fruit and its preparation Sanlindan.The samples were extracted with ether under reflux. The ether extract was evaporated to dryness and the residue was then extracted with methanol,the methanol solution was loaded onto the top of a chromatographic tube containing neutral AI2O3.then eluted with methanol.The eluate was collected and evaporated to dryness,the residue was dissolved in 5ml methanol and then spotted on a silica gel GF2s4 plate,and developed with a mixture of petroleum ether(30~60℃)0-ethyl formateformic acid(15 :5:1) for 10~12cm,The plate was scanned at 254nm(λs)and 400nm(λR)by reflection-zigzag mode. Linear calibration curve was obtained for γ-schisandrin in the range of l-5μg. The average recovery of γ-schisandrin was 99.65%.
