用地高辛标记原位杂交技术定位黄鳝rRNA基因于二价染色体3q12─q24和7q14─q26
Localization of rRNA Genes of Rice -field Eels(Monopterus albus Zuiew) on the Bivalents 3q12-q24 and 7q14-q26 by Digoxigenin Labelled in situ Hybridization
【摘要】 以地高辛标记重组质粒PTA71中所含的小麦rRNA基因作为探针,与EcoRI酶切的黄鳝核基因组总DNA经Southern杂交,呈现2条带,片段长度分别为12.8kb和4.6kb。再运用染色体原位杂交技术及杂交后多重带显带技术,定位rRNA基因于黄鳝二价染色体3q12-q24和7q14-q26两个区间,其分布位点与硝酸银染色法结果相符。此外,还讨论了在黄鳝二价体上开展基因定位研究的突出优点。
【Abstract】 Using the digoxigenin labelled wheat rDNA inserted into PTA71 as the probe,two bands,12.8kb and 4.6kb, have been shown to hybridize with total DNA of Rice-Field Eels digested with EcoRI by Southern blotting.Two rDNA locus have beenmapped on bivalents 3q12-q24 and 7q14-q26,respectively, by in situ hybridization combined with two multiple banding methods after hybridization.The results havebeen confirmed by Ag-NOR staining. The apparent advantages of mapping genes onbivalents have been discussed in detail.Received November 8, 1993.
