【摘要】 本文报道鸡多次内服（３０ｍｇ／ｋｇ，每隔１２ｈ１次，连用６次）及单剂量内服（１２０ｍｇ／ｋｇ）喹乙醇在肝、肾、肌肉及脂肪４种组织中的消除及残留的研究。用乙睛－乙酸乙酯（３／２，Ｖ／Ｖ）提取组织中的药物，反相高效液相色谱法测定组织中喹乙醇的浓度。色谱条件：Ｃ＿（１８）－ＯＤＳ柱（２５ｃｍ×４．６ｃｍ）；流动相为甲醇－水（１５／８５，Ｖ／Ｖ）；流速１．０ｍ／ｍｉｎ，紫外检测波长２６０ｎｍ。鸡多次内服喹乙醇，于第６次给药后在肝、肾、肌肉及脂肪中药物的消除半衰期分别是４．３２、４．５４、３．３１及３．０３ｈ。若规定组织中喹乙醇的最高残留限量０．００１ｍｇ？ｋｇ为可食用，则休药期应为２．５ｄ。内服单剂量喹乙醇（１２０ｍｇ／ｋｇ）后在肝、肾、肌肉及脂肪中药物的消除半衰期分别是１６．１６、１５．１９、１２．３７及１１．００ｈ，若规定食用组织中喹乙醇的最高残留限量为０．００１ｍｇ／ｋｇ，则休药期应为８．５６。结果表明，鸡内服中毒剂量的喹乙醇（１２０ｍｇ／ｋｇ）显著减慢其在体内的消除，而延长在组织中的消除半衰期及残留时间。更多还原

【Abstract】 Elimination and residues of olaquindox in chicken tissues were studied after lnulti-ple oral administration (30mg/kg,every 12h for 6 doses)and single oral administration(120mg/kg). Olaquindox in tissues(liver,kidney, muscle and fat)was extracted withacetonitrile-ethyl acetate(3/2,v/v).The tissue concentrations of olaquindOx weredeterminated using reversed-phase high performance liquid chromatography (RP-HPLC)with the following conditions:The mobile phase consisted of methanol-deionized water(15/85,v/v),and flowed at a rate of 1. 0ml/min,wavelength of UV detector was 260nm.Elimination half-life of olaquindox in liver,kidney, muscle and fat was was 4. 32,4.54, 3.31 and 3.03h, respectively,after the sixth dose of multiple oral administration(30mg/kg).WithdrawI time was 2.5 days if the above fou,tissues with 0.001mg/kgolaquindox were not consumed for human.Elimination half-life of olaquindox in liver,kidney, musele and fat was 16.16, 15.19, 12.37 and 11.05h, respectively,after singleoral administration(120mg/kg).Withdrawl time was 8.5 days if the above four tissueswith 0.00mg/kg olaquindox were not consumed for human。The results show that thetoxic dosage olaquindox(120mg/kg)slowed the elimination of the drug in chicken,andthus prolonged the elinination half-life and residue time of olaquindox in chickentissues.更多还原
