新型空气热循环式PCR仪的实验室鉴定及应用
【摘要】 北医三院医工处研制出新一代空气热循环式PCR仪,其特点为:光源加热,空气传导,冷风降温,玻璃毛细管为反应容器,因此一个PCR循环时间大大缩短。快速PCR能保持Taq酶活性,增加PCR产量,减少非特异性扩增。30个PCR循环最短可在30分钟完成(反应体积5-10μL),当反应体积为50-100μL,时,30个PCR循环可在90分钟完成。经对结核杆菌、乙型肝炎和丙型肝炎病毒进行PCR检测,鉴定此仪完全达到设计要求,可广泛应用于临床检测及目的DNA的制备,性能达到国际同类产品水平,填补了国家空白。
【Abstract】 A new Air Thermo-Cycler war made in The Third Affiliated Hospital of ’Beijing Medical College featuring lamp heating,air conducting and wind cooling,The vessel of PCR reaction is gIass capillary,so the time of orie cycle is shortened,and the rapid reaction keeps the high activity of Taq DNA polymerase. The result of test for Tuberculomyces, HBV and HCV with the PCR cycler shows that:not only can it be applied in examination of clinic desease,but also in amplification of genome DNA
