多药抗性逆转药物对HL60/Har细胞内钙调素水平的影响

【摘要】 利用显微分光光度术测定细胞内钙调素及钙活化钙调素的含量。2μMEBB、0.5μMCSA,15μMTFP作用HL60/Har细胞18h后EBB显著降低细胞内钙活化钙调素水平(P<0.001),对总钙调素影响不大;CSA对二者都有抑制;TFP强烈抑制细胞内总钙调素水平,但对钙活化钙调素影响较小。结合EBB、CSA、TFP作为逆转肿瘤细胞多药抗性药物的事实,我们推测肿瘤细胞多药抗性的形成及抗性逆转与细胞内钙调素和钙活化钙调素水平有关。

【Abstract】 EBB, CSA ahd TEP can reveree the resistance to antitumor drugs of HL60/Har. For elucidation of the mechanism, we examined Ca2+ activited CaM (Ca2+-CaM) and total CaM level in HL60/Har cells. Theresults showed that 2μM EBB could dimmish the total Ca2+-CaM significantly, but it did not decrease thetotal CaM. Cells treated with 15μM TEP had a low level of total CaM and had a slight effect onCa2+-CaM by 0.5μM CSA. The results suggest that the overcoming of multidrug resistance of HL60/Har isassociated with the reduction of CaM and Ca2+-CaM in cells.
