香茶菜黄酮诱导人肝癌细胞系GHC-3恶性表型的逆转
INDUCTION ON THE MALIGNANT PHENOTYPICAL RE- VERSION OF HUMAN HEPATOCELLULAR CAN- CER CELL LINE (GHC-3) BY RLF IN VITRO
【摘要】 香茶菜提取物黄酮体外对人肝癌细胞系GHC-3的诱导,从细胞生长特性、膜受体、肿瘤标志物、癌基因产物和遗传性状等方面观察到细胞恶性表型的逆转。经过药物作用10周后发现细胞形态由多边形上皮样逐渐转变为梭形,生长速度减慢,分裂指数、集落形成、ConA凝集和ConA受体、DNA含量及~3H-TdR掺入均明显减少,染色体分布较集中,出现二倍体细胞等变化。进一步以三种单克隆抗体测试,AFP和ras P21合成以及荧光标志抗体显示的膜抗原都明显减弱。这些改变在脱离药物后一个月以上仍维持稳定。以上结果表明,香茶菜黄酮可诱导人肝癌细胞改变恶性表型特征,促使向正常或降低恶性方向逆转。
【Abstract】 The changes of human hepatocellular cancer cell line (GHC-3) induced by Chinese herbal medicine Flavones from Rabdosia (RLF) was studied in vitro. The malignant phenotypical reversion of GHC-3 cells was observed such as the properties of cell growth, cancer cell membrane receptor, tumor marker, oncogene products and inheritable character and so on. It was found that the treated cell with RLF for 10 weeks not only gradually changed morphogenesis from the polygonal epithelial-like cells into the spindle cells., but also remarkably decreased in so far as their growth speed rate, mitotic index, colong formation, Con A agglutination and Con A receptor, DNA amount as well as total incorporation of 3H-TdR into cells and the treated cells which have the chromosomes seemed to be collectively and sonic diploid cells were counted.Furthermore, the treated cells were assayed with G3M-McAb, AFP-McAb and ras p 21-Mc-Ab. The results show that AFP or ras p 21 synthesis decreased gradually and the antigen of GHC-3 on the cell membrane by the fluorescence label of antibody became weeker.After transferred to standerd medium more than one month, the induced biological properties of the treated cells were continued. These results mentional above suggest that RLF was able to change GHC-3 cell’s malignant phenotypic characteristics and produce a reversed alteration.
