用6个小麦─黑麦衍生系筛选与黑麦抗条锈病基因连锁的RAPD标记
RAPD MARKERS FOR RYE CHROMATIN IN SIX WHEAT-RYE DERIVATIVES RESISTANT TO STRIPE RUST
【摘要】 用RAPD分析6个农艺性状不同的小麦─黑麦抗条锈病衍生系材料,建立与黑麦抗条锈病基因连锁的分子标记。结果表明,所用6个随机引物中,4个引物在6个衍生系中扩增出6个黑麦特有的产物,其大小约197~980bp,分别被标记为J17,J18a,J18b,J11a,J11b和D5。其中,J17、J11b和D5标记在6个衍生系中都存在,是与黑麦抗条锈病基因连锁的RAPD标记;J11a在4个衍生系中存在;J18a和J18b在3个衍生系中存在。表明这6个衍生系可能涉及3~6个黑麦片段的易位,是不同于1B/1R类型的一批新的小麦─黑麦异易位系。结果还表明,RAPD分析在小麦遗传背景中检测异源染色体片段转移,是一项有效的技术,有望成为早代选择异易位系的一个分子指标。
【Abstract】 Detection of rye gene for resistance to stripe rust using RAPD marker was performed using sixwheat-rye derivatives. Four out of skprimers yielded six sPecific products of rye(J17,J18a,J18b,J11a,J11b and D5),0f which J17,J116 and D5 markers have been identified in all of six deriva-tives,J11 a in four derivatives , J18 a and J 18b only in three derivatives.RAPD assay for identify-ing rye or alien DNA fragment in wheat genetic background is suggested as a way to isolate newtranslocation in selection of early generation.Furthermore,in studying on alicn gene,effectof envi-ronment could be induced,breeding procedure and selection efficiency could be improved.
