抗口腔支原体、精氨酸支原体单克隆抗体的制备及其应用方法的初步研究

【摘要】 用口腔支原体、精氨酸支原体抗原免疫BALG/c小鼠,取其脾细胞,与小鼠骨髓瘤细胞(Sp2/0)融合,获得了7株稳定分泌抗口腔支原体单克隆抗体(McAb),2株分泌抗精氨酸支原体McAb的杂交瘤细胞株。间接ELISA测定其抗体效价可达1:103~1:107;9株McAb中1株为IgG2a,其余均为IgG1。初步建立了ELISA检测支原体的方法。

【Abstract】 Monoclonal antibodies(McAbs) were prepared by fusing of Sp2/0 cells with spleen cells of BALB/c mice immunized with the antigens of M.orale and M.arginine respectively.Antibodies in hybrid culture supernatants were detected by ELISA,and positive hybrids were cloned by the technique of limiting dilutions.Nine strains of hybridoma cell lines have been obtained,of which seven strains secreted anti-M. orale antibodies and two strains secreted anti-M.arginine antibodies.One of the immunoglobulin class/subolass of McAbs was IgG2a and the others belinged to IgG2.ELISA was used for identifying titers of nine McAbs which were from 1:103 to 1:107.The methods,which were used to detect M.orale by sandwich ELISA and to detect M.argininge by competitive ELISA,have been established.
