耐碱性芽孢杆菌碱性淀粉酶的研究──发酵试验与酶的性质
Study on Alkaline Amylase from Alkatophilic Bacillus Megaterium ──The Conditions for Enzyme Preduction and Enzymic Propernes
【摘要】 本文报道碱性α-淀粉酶14L罐发酵工艺及其酶的性质。发酵过程中,当搅拌转速为400r/min,通风比为1:1.25L/(L·min),发酵周期44h,酶活力可达740u/ml。采用有机溶剂沉淀法提取酶,回收率可达90%。Ca ̄(2+)对酶活性无影响,EDTA对酶有激活作用。酶样保存半年,活力损失小于15%。
【Abstract】 he fermentation of alkaline amylase from the alkalophilic Bacillus megateriun L-49 in 14L tank has been studied. The optimum ventilation quantity and agitation speed were 1:1. 25 vvm and 400r/min respotivily.The fermentation cycle was 44h and the highest alkaline amylase activity was 740u/ml.The enzyme was recovered by precipitation method of organic solvents.The effects of surfatants,EDTA and Th2+ ion on enzymes activity were also studied.
