【摘要】 通过体外ＤＮＡ拓扑异构酶Ⅱ介导ＤＮＡ裂解实验观察了芦笋提取物对该酶活性的影响。从小鼠白血病Ｌ１２１０细胞中提取ＤＮＡ拓扑异构酶Ⅱ，以ｐＢＲ（３２２）ＤＮＡ为底物，将１μｇＤＮＡ与０．２Ｕ拓扑异构酶Ⅱ温孵，当分别加入芦笋提取物２２０、２２、２．２μｇ／ｍｌ３个终浓度时，裂解能力可提高到原来的５倍，说明该药的直接作用靶点是ＤＮＡ拓扑异构酶Ⅱ，而不是直接裂解ＤＮＡ。更多还原

【Abstract】 The purpose of the test is to investigate the principle of activity of Extractum of Asparagus Officinalis promoting DNA topoisomerase Ⅱ activity of cleaving DNA. DNA topoisomerase Ⅱwas isolated from mouse leukemia L1210 cells and took PBR322 DNA as the substrates. After incubation of pBR322 DNA 1μg with 0.2U of topoisomerase Ⅱ, the presence of Extractum of Asparagus Officinalis by 220;22; 2.2μg/ml respectively, it reproducible stimulated DNA cleavage by 5-fold. It may be the fact that DNA topoisomerase Ⅱ is a direct target in antitumor activity of Extractum of Asparagus Officinalis.更多还原
