备用根手术对背角组织促神经突起生长活性的影响
The Effect of Spared Root Operation on Promoting Neurite-Outgrowth Activity of Spinal Dorsal Horn
【摘要】 将5只单侧后肢备用根术后5天猫的背角组织提取液用于培养鸡胚背根节及电泳分析。手术组(手术侧)提取液大于50KD组份培养的背根节的神经突起明显长于非手术组(非手术侧)者,提示手术组此组份的促神经突起生长活性显著增强;该组份各电泳带的吸收光谱显示,手术组迁移率为0.1的蛋白带的吸收峰面积百分比明显增大,而迁移率为0.5的蛋白带的吸收峰面积百分比则明显减小,表明手术组提取液大于50KD组份中此两带的蛋白质含量分别有所增减。推测可能系备用根手术导致提取液大于50KD组份中某一促神经突起生长活性物质含量增加,而另一抑制神经突起生长物质含量减少,从而导致背角组织促神经突起生长活性的增强。
【Abstract】 Five days after unilateral L1-S2 partial dorsal rhizotomy (L6 as spared root) of five adult cats. the dorsal horn of both sides from their spinal T13-S3 were taken separately.The extract from dorsal horn was superfiltered into two fractions:a smaller than 50KD in molecular weight(MW)fraction and larger than 50KD in MW fraction.The two fractions of both sides were used to culture dorsal root ganglia(DRG)from chick embryo with a modified hanging drop method. The average neurite length of DRG was measured at 48h and the extract was analyzed with polyacrylamide gel electrophoresis.The results were as follows:1.The average neurite length of DRG cultured with smaller than 50KD fraction of operated side was no statistically difference from control side(P>0.05).2.The average neurite length of DRG cultured with larger than 50KD fraction of operated side was significantly longer than that of the control side(P<0.05).These results show that the promoting neurite-outgrowth activity of the larger than 50KD fraction of operated side increased obviously,but the one smaller than 50KD was not.3.The areal percentage of absorption Wave in two protein zones of the larger than 50KD fraction of the operated side changed significantly:one(relative mobility,RM0.1)increased,the other(RM0.5)decreased(P<0.053).This indicates that the protein content of the two zones of the operated side changed.In other words,protein content of RM0.1 increased, and that of RM0.5 decreased.The author suggests that the protein of RM0.1 zone may involve in promoting of neurite-outgrowth and the protein of RM0.5 zone may involve in inhibiting of neurite-outgrowth.Therefore,the promoting neurite-outgrowth activity of dorsal horn in creased after partial lumbosacral dorsal rhizotomy.
