侵染西番莲的CMV分离物的研究
A CMV ISOLATE INFECTING PASSIONFRUIT
【摘要】 从海南西番莲病株中分离获得一个病毒分离物,根据鉴别寄主生物学反应、粒体性质和血清学性质鉴定为黄瓜花叶病毒(CMV)。该分离物接种西番莲可引起花叶和叶下卷,有时可变成隐症,在一些指示植物上的症状和其它CMV株系相同,但在三生烟接种叶上表现局部黄斑,然后逐渐发展成系统花叶。提纯获得了纯化病毒,并免疫家兔制备出抗血清。电镜观察病毒粒体为等轴对称形态;SDS-PAGE显示其外壳蛋白由一种分子量为28KD的亚基组成;琼脂免疫双扩散和ELISA实验结果表明,该分离物和CMV有密切的血清学关系。
【Abstract】 A virus isolate identified CMV on the bsis of host reaction ,particle morphology,serolo-gy,was obtainedfrom passionfruit(Passiflora eddisf.flavicarpa)growing in Hainan,This iso-late could induce mosaic and leaf curling,or in some cases latent symptoms of the passionfruit.Bi-ological reaction of this isolate on some major indicator plants was found identical to that of otherCMVisolates,but differences were observed On Nicotiana tabacum var,Sumsun which showedlocal yellow spot on inoculcqted leaves followed by gradual development of systemic mosaic symp-tom。The virus was purified and antiserum was prepared by immunized rabbit.Electron mi-croscopy found the virus possessing isometric particle。SDS-PAGE indicated the viral coat proteinbeing composed of 28 KD subunit.Agarose gel double diffusion test and ELISA indicated that thevirus isolate infecting passionfruit was serologically closely related to CMV。
