共焦显微镜光学切片成像定位花粉管的细胞骨架成分
Localization of the Cytoskeleton Composition in the Pollen Tube by Imaging of Optical Sections with CLSM
【摘要】 运用共焦激光扫描显微镜(CLSM)对花烟草(NicotianaalataLinketOtto)花粉管荧光标记样品进行连续光学切片,并进行整个样,吊成像和局部成像,结果显示出,被动蛋白(ki-nesin)抗体标记的囊泡状物主要分布在花粉管顶端约20μm范围内、近质膜2μm的区域;被罗丹明一鬼笔环肽(Rhodamine-phalloidin)标记的微丝则分布比较广泛,但光学切片显示出花粉管顶端中央有一个长约25μm的区域不被标记。结果表明,CLSM光学切片成像,是光学显微水平定位细胞成分的有效方法。
【Abstract】 Confocal Laser Scanning Microscope was used to localize kinesin and microfilaments in the pollen tubes of Nicotiana alata. The results from the optical sections and the images of CLSM showed that the vesicles were recognized by antikinesin antibody and they were distributed in the area about 2 μm to the plasma membrane around the tip of the pollen tube,and the microfilaments labelled with Rhedamine-phal-loidin were distributed widely,but an area about 25μm long in the pollen tube tip was not labelled.The results indicate that Imaging of Optical Sections with CLSM is a good method to localize the cytoskeletoncomposition at the level of Optical microscopy,
