枯草杆菌B_3抗植物病原真菌蛋白的纯化及其性质的研究

【摘要】 枯草杆菌菌株B3(Bacillussubtilis)用NB培养液振荡培养(30℃,110r·min-1pm,60h),离心(5000r·min-1,15min,4℃)去除菌体,上请液经超滤浓缩与离子交换层析,分离并纯化出一种抗植物病原真菌蛋白,经HPLC鉴定纯度为98.0225%。电泳分析该蛋白分子量为12882.5D,等电点为4.8。氨基酸组成分析含13种氨基酸残基,其中谷氨酸含量为最高,甘氨酸、半胱氨酸、苯丙氨酸和精氨酸含量则较低。纯化后的抗菌蛋白对小麦赤霉病菌(Fusariumgraminearum)和小麦纹枯病菌(Rhizoctoniacerealis)有很强的抑菌作用,有效浓度分别为31.25,62.5mg·kg-1。

【Abstract】 The NB culture of B3 was centrifuged and the supernatant wasconcerntrated by ultrafiltra-tion,A kind of antifungal protein was characterized from the concerntrate by DEAE一Sepharose F. F col-umn chromatography, FPLC superose一12 gel filtration and monoQ column chromatography.Its puritywas identified to be 98.0225%by HPLC. The molecular weight and isoelectric point were determined tobe l2 882.5 daItons and 4. 8 respectively by autornated electrophoresis. An1mino acid analysis result indi-cated that it consisted of l3 ammino acids,and the content of Glu ls the hightest. Characterized antifungalprotein was greatly inhibitory to Fusarium graminearum and Rhizoctonia ceieatis,and the effective con-cerntration was 31.25 mg· kg-1 and 62.5mg· kg-1,respectively.
