反转录-多聚酶链反应扩增小鼠睾丸17α-羟化酶mRNA
The emplification of 17-α-lydroxylase with reverse PCR
【摘要】 17α-羟化酶催化雄激素合成的中间步骤,它对了解睾丸间质细胞内雄激素的合成具有重要的意义。本研究首先提取小鼠睾丸组织RNA,然后以17α-轻化酶DNA两端的特定序列为引物,反转录合成cDNA,再以cDNA为模板行PCR扩增cDNA。结果显示,反转录PCR后获得一条360bp的电泳带,为17α-羟化酶cDNA。结果表明,用反转录PCR方法扩增睾丸17α-羟化酶mRNA是切实可行的。
【Abstract】 The synthesis of androgen is catalysised by 17-α-lydroxylase. The study of 17-α-lydroxylase mRNA could improve the understanding of this procedure in Leydig’s cells. In this study,mouse testis RNA was isolated at first. Then specific nucleic sequences at the end of 17-α-lydroxylase DNA were used as primer to synthesis cDNA with 17-α-lydroxylase mRNA as templet; At last,PCR was used to emplify cDNA, The result showed that a 360bp cDNA was obtained after PCR was useful for the empilification of l7-α-lydroxylase mRNA.
