LEIA检测人型支原体抗原方法的研究
LUMINESCANT ENZYME IMMUNOASSAY FOR MYCOPLASMA HOMINIS ANTIGEN
【摘要】 探讨了邻苯三酚-H2O2发光系统的最适实验条件。结果表明,发光溶液的最适pH为6.5~7.0,邻苯三酚浓度为0.3%,H2O2浓度为0.45%,测定前需37℃暗适应15min。发光强度用DG3030发光仪检测。该法测定人型支原体抗原的敏感度为1.02ng/ml。用本法与ELISA对51份临床阴道分泌物标本平行测定,结果没有显著差异。
【Abstract】 uminescant Enzyme Immunoassay(LEIA)was established for detection of Mycoplasma ho-minis antigen with HRP-Pyrogallol-H2O2 system,It is found that pH 6.5~7.0,0. 3%pyrogallol,0.45%H2O2 are suitale conditions for the assay.Incubation under 37℃ for 15 minutes before test-ing chemiluminescence value is necessary for stablizing the results. The sensitivity for M. hominisantigen is 1.02 ng /ml.Results of 51 clinical specimens with LEIA were not significantly differentfrom those with ELISA. It is suggested that LEIA be a usuful method for epidemiologic study ofMycoplasma hominis infection as well as ELISA.
