【摘要】 用上海医科大学组胚教研室制备的大鼠肝抑素粗制品,半纯品和纯品,以～3H-TdR掺入液闪测定(CPM)法,检测它们对分离培养的再生肝肝细胞增殖的半数抑制量.结果表明:三级样品的半数抑制量分别为466.62μg/ml,23.44μg/ml和0.2333μg/ml,肝抑素半纯品的生物活性是粗制品的20倍,纯品是粗制品的2000倍.用放射自显影法验证肝抑素三级样品对再生肝细胞摄取～3H-TdR的影响,也表明可使再生肝肝细胞核标记率明显下降,并与液门测定结果一致,以肝抑素纯品的活性最强.结果证明提取的大鼠肝抑素纯品具有抑制再生肝细胞增殖的高度活性.更多还原

【Abstract】 The crude, partially purified and purified products of rat hepatic chalone(HC) were prepared in our laboratory and their 50% inhibiting dose(ID50) to regenerating hepatocytes was assayed in vitro by means of ’H-TdR incorporation (Fluid scintillation, cpm). The results showed that the ID50 of the three HC products was 46662ug/ml, 2344ug/ml and 02333 ^g/mlrespectively.The biological activities of the partially purified HC and the purified HC were 20 and 2000 times stronger than that of the crude product respectively. The effects of the three HC products on the ’H-TdR uptake of the regenerating hepatocytes were also verified by autoradiography. The results were consistant with those assayed by fluid scintillation. The activity of the purified HC is the strongest is all the three products.更多还原
