高产普鲁兰菌株的筛选和发酵条件探索
screening and Fermentation of High Production Strain of Pullulan
【摘要】 通过对茁芽短梗霉As3.2756四次紫外诱变,获得了一株高产普鲁兰的变异株PB518,使糖的转化率达55%以上,经多次传代表明该菌株稳定。同时考察了该变异株的发酵条件,最适条件为起始pH6.0,接种量3.0×10 ̄8个/mL通气量为培养基占摇瓶体积的1/10,发酵温度28℃,发酵时间96h。
【Abstract】 The high-yield pullulan produced by mutant PB 518 of Aureobasidium Pullulans As 3.2756was isolated after treating the parent strain with ultraviolet.Its high productive character remains sta-ble after several generations The optimum conditions have been studied and confirmed as following:sucrose was 50 g/L,(NH4)2SO4 0.4 g/L,KH2PO4 2 g/L,MgSO4·7H2O 0.1g/L,NaCl 2 g/L,yeast extract 2 g/L,ini-tial pH 6.0(before sterilizing),the amount of inocula 3×10 ̄8/mL, the temperature 28 ℃ the time 96hours. The conversion of pullulan is above 55%.
