细胞间粘附分子在脑血管内皮细胞的表达

【摘要】 本文用细胞培养方法,观察脑血管内皮细胞粘附分子(ICAM-1)的表达和内皮细胞与血液单核细胞的粘附作用。结果显示,10u/mlTNF-α即可使内皮细胞ICAM-1表达增加,自发性高血压大鼠(SHR)增加的程度大于正常血压大鼠(WKY)。干扰素(IFN)和白细胞介素1(IL-1)也可使ICAM-1表达增加。高浓度细胞因子均使SHR和WKY内皮细胞ICAM-1表达增加,但二者之间无明显差别。经LPS或IL-1和TNF-α刺激的SHR内皮细胞与单核细胞的粘附率明显高于WKY。结果证实,TNF-α,IL-1和IFN可使SHR和WKY脑血管内皮细胞ICAM-1表达和单核细胞粘附增加,两组动物细胞相比,SHR内皮细胞ICAM-1表达及粘附率增加较多,提示SHR脑血管内皮细胞可能对细胞因子的敏感性升高。

【Abstract】 ell adhesion plays a very important role in many pathological processes. The expression ofICAM-1 on cultured cerebrovascular endothelial cells and the adhesion reaction of the endothelialcells with monocytes in vitro were observed in this experiment. The results showed that 10u/mlTNF-α increased more expression of ICAM-1 in SHR endothelial cells than that in WKY. Highconcentration of cytokines increased the ICAM-1 to a higher level in both SHR and WKY, but nosignificant difference between them. Comparison of adhesion reaction, the ability of freshly iso-lated peripheral blood monocyte to adhere to cerebrovascular endothelial cells treated with LPS orIL-1 and TNF-α was significantly greater in cell cultures from SHR than from WKY. These datademonstrate the increased expression of ICAM-1 on endothelial cells from SHR. Also, the in-creased snesitivity of endothelial cell cultures to cytokines was related to hypertension.
