【摘要】 用人Ａ型及Ｂ型红细胞分别免疫ＢＡＬＢ／Ｃ小鼠，取免疫脾细胞与同系ＳＰ２／０骨 髓瘤细胞融合，经克隆筛选出８株抗Ａ和５株抗Ｂ杂交瘤细胞株，用小鼠腹水单克隆抗体分 别做凝集试验。亲和力测定、标本检测及ＣＰＶ、ＰＨＡ、ＣｏｎＡ对单克隆抗体效价的影响。结果表 明除Ａ４和Ｂ４株外，其余各株凝集效价均在１：１２８０以上；亲合力与抗Ａ、抗Ｂ血清相似；标本 检测符合率为１００％。结果提示：用含ＰＨＡ、ＣｏｎＡ、ＣＰＶ的杂交瘤细胞注射小鼠，所产生的单 克隆抗体效价比单独用杂交瘤细胞产生的单克隆抗体均高出２倍。更多还原

【Abstract】 Eight strains of hybridoma cell secreted anti human blood group A monoclonal antibody and five strains secreted anti B. They were cloned by fusing SP 2/0 myeloma cells and spleen cells of BALB/C mice,which were routinely immunized with human blood group A and B RBC. The hybridoma cells alone or with ConA PHA CPV were injected into the ascitic cavity BALB/C mice,and the ascites were collected and characterized by agglutination test,affinity test and sample assay. The results showed that the titers of the ascites of all strains except A4 and B4 were above 1: 1280. The affinities were similar with standard anti blood group A and B sera,and the samples tests were also exactly conincident. The titers of ascites were two times as high as hybridoma cell alone,if hybridoma cell with PHA,ConA and CPV were injected.更多还原
