烟草盐适应愈伤组织生理特性的研究

【摘要】 采用逐步提高NaCl浓度的方法,获得了耐1%NaCl的烟草(NicotianaristicaL.cvGansuYellow一flower)愈伤组织变异系(S_1)。与原始型愈伤组织(S_0)相比,在1%NaCl胁迫下,S_1的干重增量高于S_04.6倍,组织渗透势下降,而含水量提高;游离脯氨酸、蔗糖、还原糖含量及乙烯释放速率的增加S_0高于S_1,但腐胺的累积量相反。S_1对甘露醇(Manitol)渗透胁迫和Na_2SO_4盐胁迫也具有适应性,但对低温(0一1℃)敏感。ABA促进了S_0对盐胁迫的抗性,但对已耐盐的S_1无类似作用。

【Abstract】 A 1%NaCl一tolerant cell line has been selected from tobacco callus(Nicotiana ristica L. cv Gansu Yellow一flower)by stepwise increases in NaCl concentration in the medium. Resistance showed stability through 3 generations in the absence of NaCl. In the presence of 1%NaCl,the selected cell line(S1)had higher water content but lower os- motic potenial,less free proline,sucrose and reducing sugars content than the nonselected cell line(S0).The selected cell line responded to salt stress(1%NaCl)by accumulating markedly more Put and deducing the rate of ethylene evolution than did the nonselected cell line,Which im- plied that the cells were bound for salt一adaptation through the selecting procedure. The NaCl一selected cell line(S1)grew better under both Na2SO4 and mannitol stress than the nonselected cell line(S0),but the susceptity to the low temperature(0一1℃)was reverse. 10 ̄(-5) M ABA stimuiated the growth of the nonselected cell line in the medium with 171 mM NaCl or 85.5 mM Na2SO4,but exhibited no such effect on S1 and S0grown on the nonionic osmotic solutes(342 mN mannitol or 342 mM sucrose).These results suggest that ABA is involved in adaptation of cells to salt,and is not effective in promiting adaptation to water deficits elicited by nonionic osmotic solutes.
