钙黄绿素蓝荧光法测定食品中的微量锌(Ⅱ)

【摘要】 本文研究了在CTMAB存在下,用钙黄绿素蓝荧光法测定食品中的微量锌(Ⅱ)的实验条件。在本实验条件下,Zn(Ⅱ)与钙黄绿素蓝生成1:1的配合物使钙黄绿素蓝的荧光增强,最大激发波长和最大发射波长分别为353nm和440nm。锌量的线性范围为0.017~0.70μg/10mL,检测限为1.7ng/mL,标准曲线的中间部分的相对标准偏差为1.56%。方法用于测定脱脂奶粉、大米粉、含锌营养食盐中的微量锌(Ⅱ),相对标准偏差分别为2.59%、4.60%和0.49%。

【Abstract】 In this work,a new spectrofluorimetricmethod for the deterrnination of zinc(Ⅱ)in foodstuff with calcein blue(CB)was devel-oped. The formation of CB-Zn(Ⅱ)complexleading to the enhanceinent of fluorescenceintensity of CB was applied to the determi-nation of Zn(Ⅱ).The addition of CTMAB(cetyltrimethyammonium bromide)to the solution of CB-Zn(Ⅱ)complex produces anincrease of fluorescence intensity of CB andan increase of stability of the Zn(Ⅱ)-CBcomplex. The maximum excitation and e-mission wavelengths of CB reagent blankare 332 and 440nm and those of the CB-Zn(Ⅱ)complex are 353 and 440nm,respec-tively, The concentration range for a linearrelationship is 0. 017~0.70μg/10mL andthe detection limit is 1. 7ng/mL for Zn(Ⅱ).The analytical results indicate a precision of1. 56%at the middle region of the calibra-tion curve,This method is simple and selec-tive for Zn(Ⅱ)and it has been applied tothe determination of Zn (Ⅱ)in foodstuffwith satisfactory results.
