过量硒对培养心肌细胞的毒性作用

【摘要】 于体外培养大鼠心肌细胞培养液中添加亚硒酸钠,观察到当硒的浓度增至0.5~1.0×10-6时,心肌细胞[3H]—TdR(脱氧胸苷)掺入降低,细胞内肌酸激酶漏出增多,细胞极度收缩,表面平坦失去微绒毛结构,细胞膜破损、线粒体肿胀、肌浆网扩张、肌丝结构消失。表明硒过量能引起心肌细胞的直接损伤。

【Abstract】 The effect of sodium selenite addition, especially the cytotoxicity of its over-dose on cultured neonatal rat heart cells was investigated. The results showed that over-dose selenium (0. 5-1. 0ppm) could decrease [3H] -TdR incorporation into DNA and increase the leakage of intracellular creatine phosphokinase in the cultures. Excessive selenium could also cause the ultrastructure changes, e. g. disorder of cellular contraction, rupture of cellular membrane, swollen of m’itchondria, dilation of sarcoplasmic reticulum and disappearance of myofilament, etc. These suggested that excessive selenium might directly cause the damage of myocytes.
