【摘要】 本文采用 4.2mol/L异硫氰酸胍、25mmol/L柠檬酸钠、0.1mol/L2一巯基乙醇,0.5%（W/V）十二烷基肌氨酸钠作为裂解液进行一步法程序式抽提血清标本HBV—DNA模板用于多聚酶链基因扩增.该法与常规DSD抽提法及微量直接法进行比较,具有方法简单、稳定、不易污染且敏感性高.抽提过程可在1个小时内完成,经酶切及a³²P标记探针放射自显影鉴定PCR产物具良好特异性,该法尤其适应于大量临床标本的检测,值得推广使用.更多还原

【Abstract】 A new method of HBV - DNA isolation by a simple extraction with an acid guanidinium thiocyanate -phenol saturated with water in PCR was described. The method was simple and can be completed in 1hour. Being compared with other HBV-DNA isolation methods ,this technique had a higher sensitivity, and was particular useful in processing large number of clinical samples.更多还原
