香蕉花叶病的酶联免疫检测技术研究
ON THE ELISA METHODS FOR DETECTING BANANA MOSAIC DISEASE
【摘要】 用分离自香蕉病株的黄瓜花叶病毒香蕉毒株(CMV-B)和烟草花叶病毒香蕉毒株(TMV-B)制备抗血清,其效价As-CMV-B为1:5000,As-TMV-B为1:8000。用这两种抗血清检测香蕉花叶病,间接ELISA和PAS-ELISA法可检测香蕉病叶汁液的最大稀释度均为1:1280,健叶汁液无非特异性反应,Dot-ELISA可检测病叶汁液的最大稀释度为1:640,健叶汁液在稀释度为1:20时有轻度非特异性反应。用间接ELISA法检测了田间香蕉病株和香蕉试管苗标样共158份,其中CMV的带毒率为58%,TMV的带毒率为19%,二者复合感染率为8.5%。
【Abstract】 The mice antisera of the cucumber mosaic virus-banana isolate (CMV-B) and tobacco mosaic virus-banana isolate (TMV-B) were prepared. The titeres of the two antisera, which were tested by indirect ELISA, were 1:5000 and 1: 8000, respectively. The banana mosaic disease were detected with the two antisera. The leaf juice of banana plant with mosaic disease is readily reacted postively with the antiserum by indirect ELISA when it was diluted even to 1:1280, and the leaf juice of the healthy banana plant reacted negatively. The results detected by PAS-ELISA were the same. However, the results detected by Dot-ELISA showed that the leaf juice of banana plant with mosaic disease reacted positively only before diluted to 1: 640, and the leaf juice of healthy banana plant appeared a’ little nonspecial reaction before diluted to 1 : 20.The 158 samples of banana mosaic disease from plants in the fields and the tube seedlings were detected by indirect ELISA. The results showed that samples infected by CMV and TMV were 58% and 19%, respectively, and samples infected by both of CMV and TMV were 8. 5%.
