纯化弓形虫速殖子的两种物理法探讨
INQUIRIES OF TWO PHYSICAL METHODS ON THE PURIFICATION OF TACHYZOITES OF TOXOPLASMA GONDII
【摘要】 采用纤维素粉过滤法和多聚磷酸纤维薄膜过滤法,对人工感染的小鼠腹腔液中的弓形虫速殖子进行细胞清除率和回收率比较。前者白细胞平均清除率95.4%,红细胞平均清除率90.1%,虫体回收率33.1%;后者白细胞清除率81.6%,红细胞清除率80.5%,虫体回收率48.6%。在红、白细胞清除率上,纤维素粉过滤法优于薄膜过滤法,在虫体回收率上后者略优于前者。
【Abstract】 The methods of cellulose filttation (CF) and polycarbonate filters (Poly C) have been used for removing contaminating host cells from mouse peritoneal exudates infected with Toxoplaslna gotldii tachyzoites.,We compared these two methods in purification and recovery of Toxoplasma gondii tachyzoites. Results showed that the mean removing rates of mouse leucocytes and erythrocytes were 95.4 and 90. 1 percent respectively, and recovery rate of tachyzoites was 33. 1 percent with the CF method’ The mean removing rates and recovery rate were respectively 81.6,83.5 and 48.6 percent with the Poly Cmethod. The overall results demonstrated that the removing rate of host blood cells is better seen in CF method, whereas the rccovery rate of tachyzoltes is better with the Poly C technique’
