灰稻虱体内稻条纹叶枯病毒快速检测技术研究

【摘要】 采用膜上斑点免疫结合测试技术(Dot-ImmunobindingAssay,DIBA)快速检测灰稻虱(LaodelphaxstriatilbusFallen)的毒率。试验得出用含2%牛血清白蛋白(BSA)、2%PVP、0.05%Tween20的0.02mol/LPBS作封缓覆冲液、封闭30min后使抗原(Ag)与其相应抗体(Ab)和抗体与酶标记A蛋白(HRP-SPA)各反应30min,检测效果较佳,灵敏度比相应的ELISA提高4倍。

【Abstract】 The scverity of riee stripe discase is affectal directiy by the pcrcentage of viruliferousLaodelphax strtoiellus in the field. In order to forccast the disease correctly. the percentage ofviruliferous L. strmtellus must be detected quickly and precisely. The Dotlmmunobinding Assay(DIBA) for rapid detectog the viruses in L. strmiellus was siudled it was found that the optimal bufferis 0.02mol/ L PBS containing bovine serum albunmin 2% BSA , 2%. PVP and 0.05% Tween 20, theoptimal blocking and reaction time are both 30 nuinutes. Four timee of detection senntivity are increasedby DIBA than by ELISA.
