肝性脑病大白鼠脑内血管活性肠肽受体的研究
Vasoactive intestinal peptide receptors in the brains ofhepatic encephalopathy rats
【摘要】 应用放射受体分析法对肝性脑病(HE)及对照大白鼠脑粗制突触体膜(P2膜)的血管活性肠肽(VIP)受体结合特性进行了测定。结果:HE鼠最大结合容量(Bmax)为9.56±0.29fmol/mg蛋白质,平衡解离常数(kd)为0.28±0.01nmol/L,较对照鼠Bmax值明显减少(P<0.002),而kd值差异无显著意义(p>0.20)。表明HE较对照鼠脑P2膜的VIP受体结合位点数量减少,但亲和力不变;提示其在HE发生中具有一定作用。
【Abstract】 e evaluated the effect of vasoactive intestinalpeptide (VIP) receptor, a brain-gut peptide receptor,which is capable of exciting central neurons, on thepathogenesis of hepatic encephalopathy(HE). Bymeans of radioligand binding assay, VIP receptors incrude synaptosomal membrane of rat brains were in-vestigated in a rat modei of HE induced by partialhepatectomy following carbon tetrachloride intoxica-tion and in controls. A dissociation constant (Kd) wasfound 0.28 ± 0.0lnmol / L and a maximal binding ca-pacity (Bmax) 9.56 ± 0.29fmol / mg of protein in HErats. Only decreased Bmax values were observed(P<0.002) and the Kd values were statisticallyunchanged (P> 0.20) in HE rats as compared with incontrols. The results suggest that the changes of VIPreceptors in brains play a significant role in thepathogenesis of HE. The mechanism of HE indueedby the alterations of VIP receptors in the brains wasdescribed.
