温石棉对膜蛋白构象的影响及柠檬酸铝的拮抗作用
Effects of Chrysotile on the Conformation of Membrane Protein and the Antagonistic Action of Aluminum Citrate
【摘要】 利用园二色谱技术探讨了茫棉及涞棉对人红细胞膜蛋白构象的影响及柠檬酸铝的拮抗作用。结果表明,两种温石棉在20min内,可致膜蛋白的。一螺旋量明显降低。且存在剂量依赖关系。茫棉作用较滦棉强。提示膜蛋白构象的改变是温石棉溶血作用的关键环节。经柠檬酸铝处理后,茫棉及沫棉对膜蛋白α-螺旋量的影响减弱。这为阐明柠檬酸铝拮抗温石棉细胞毒性的机制提供了重要资料。
【Abstract】 ffects of Mangai and Laiyuan chrysotile on the conformation of membrane protein and the antago-nistic actlon of aluminum citrate to them were studied with circular dichroism. Results showed that these two kinds of chrysotile could significantly reduce the amount of α-helix in membrane protein of human ervthrocvtes in 20 minutes with a dose-dependent manner Effect of Mangai chrysotile was stronger than that of Laiyuan one. It suggested the changes in the conformation of membrane protein were the kev link in chrvsotile-caused hemolysis.After treatment with aluminum citrate,these effects weakened.It provided important information for elucidating the mechanism of aluminum citrate in an-tagonism to cytotoxicity of chrysotile.
