抗氧剂在颈内动脉灌注卡氮芥时对脑的保护作用
The cerebral protecting effect of lipid su-peroxide in BCNU carotid infulsion
【摘要】 为验证颈内动脉灌注卡氮芥所致的脑损害与过氧化脂质反应的关系、及维生素C、E的联合用药效果,而建立本实验。首先取Wistar大鼠25只、参考人颈内动脉灌注卡氮芥剂量,分为五个梯度,行急行毒理实验,确定亚急性毒理实验的剂量为15mg,从而分为实验组、保护组、对照组。结果发现卡氮芥灌注侧脑组织过氧化脂质反应明显增强(P <0.001),且蛋白质总量明显减少(P<0.05),应用抗氧剂维生素C、E能有效地抑制过氧化脂质反应,预防脑毒性发生。
【Abstract】 BCNU dosage, according to the dosage of BCNU for human carotid artery infusion, was divided into five dosage gradients to perform the acute poisonous experiments by which, BCNU 15mg was dicided as dosage of subacute poisonous experiment. 25 Wester mice was divided into three groups, the first was experiment group,in which BCNU 15mg was infused into carotid artery and feeded by general diet, the second was protecting group, in which BC- NU 15mg was infused into carotid artery and then Vitamin C and E were feeded every day. the third was control group, in which isoper-meate physiological saline was infused into carotid artery without any drugs and the mice were feeded by general diet.The results show that the lipid superox-ide reaction in the cerebral tissue in the side of BCNU infusion is obviously increased, meanwhile the amount of total protein in the tissue is greatly decreased. It is demonstrated that BCNU can cause lipid superoxide reaction and make the amount of total protein decrease. The use of Vitamin C and E can effectively in-hibite the lipid superoxide reaction and reduce the poisoning.
