腺苷、谷氨酸心脏停搏液的实验研究

【摘要】 离体兔心模型,在心肌缺血120min期间,用充氧晶体及加腺苷、谷氨酸、腺苷和谷氨酸的停搏液进行灌注,比较缺血前后心肌耗氧量及心功能等指标。结果显示,腺苷或谷氨酸可通过增加有氧代谢而改善心肌保护,进而促进心脏功能的恢复。但谷氨酸部分地减弱了腺苷的心肌保护作用。

【Abstract】 Isolated rabbit heart model was used to investigate whether glutamate and/or adenosine or in combination during cardioplegic arrest could enhance myocardial protection and improve recovery from ischemia. Rabbit hearts were subjected to 120 minutes ischemia under hypothermia (5℃). Control hearts (n=6) received oxygenated crystalloid cardioplegia,and the remaining hearts received cardioplegia with either 200 μmol/L adenosine or 20 mmol/L glutamate or in combination (n= 6, respectively). The results ahowed that adenosine shortened arrest time compared with control or glutamate treated hearts, after ischemia and 30 min utes of reperfusion myocardial oxygen consumption (MVO2) was significantly higher (p<0. 05 or 0. 01),left ventricular function (DP, ±dp/dt) also much better in groups of adenosin and/or glutamate-treated hearts compared with control hearts (p < 0. 05 or 0. 01). The recovery of DP positively correlated with the increase of MVO2. Furthermore, GOT release or myocardial water content was less in adenosine or glutamate-treated groups (p<0. 05). These data suggest that adenosine or glutamate administered in cardioplegia could stimulate oxidative metabolism, maintain cell membrane integrity, and improve postischemic recovery. Glutamate partially reduced myocardial protection of adenosine.
