H2O2和ouabain对培养晶体中Na K-ATPase活性的影响及与晶体混浊度的关系
Effect of H2O2 nad ouabain on Na K-ATPase activity of cultured lenses and its relation to lenticular opacification
【摘要】 在体外培养的条件下,大鼠晶体中 Na K-ATPase 可被1mmol/L ouabain 所抑制,抑制的程度随着 ouabain 作用的时间而加强。鼠晶体被 ouabain 作用后,湿重增加,晶体膨胀,逐渐变为混浊,表明晶体中的 Na K-ATPase 在维持晶体中的离子浓度及渗透压平衡上起着重要作用。此外,通过 H2O2对培养晶体的作用,表明晶体及晶体的 Na K-ATPase 易被氧化损伤,1mmol/L H2O2 20小时即可使晶体的 Na K-ATPase 活力下降56%,致晶体混浊。
【Abstract】 Under incubation,the activity of Na K-ATPase was inhibited by 1mmol ouabain,and the degree of inhibition increased with the duration of ouabain treatment.Exposure of the rat lens to ouabain resul- ted in an increase in its wet weight with intumes- cence and gradual opacification,indicating that Na K-ATPase played an important role in maintaining the ion concentration and osmotic equilibrium of the lens.Besides,the addition of 1mmol H2O2 to the culturde lens showed that the lens and its Na K-ATP ase were damaged by oxidation in 20 hours,when the enzyme activity was reduced by 55% and the lens turned opaque.
