趋磁细菌研究—— Ⅱ.趋磁细菌WD-1的生长和磁小体的合成条件
STUDIES ON MAGNETOTACTIC BACTERIA——Ⅱ . Studies on Optimal Conditions for Culture of Magnetotactic Bacterium Strain WD-1 and Magnetosomes Formation in the Cell
【摘要】 报道了趋磁细菌WD-1生长和磁小体合成的最适条件。在pH6.7、28℃和静止培养条件下,每100mL含50~100mg柠檬酸或100~150mgα-酮戊二酸和20mg硫酸铵分别是WD-1生长的最适碳、氮源,碳氮比以5:1或4:1为好。每100mL含50mg柠檬酸、75~125mgα-酮戊二酸、75mg酒石酸或175mg乙酸钠,10mg硝酸钠或10~20mg硫酸铵,15μmol·L-1三氯化铁或硫酸亚铁分别是磁小体合成的最适碳、氮和铁源。而1~3%的氧浓度是必需的。
【Abstract】 For culture of magnetotactic bacterium strain WD-1 under the-conditions of pH 6. 7, 28℃,without shaken,optimal carbon,nitrogen and iron sources are citric acid(50~ 100 mg per 100 mL medium)or α-ketoglutaric acid (100-150 mg per 100 mL medium), (NH4)2SO4(20 mg per 100 mL medium) and FeCI3(25 μmol · L-1) ,FeSO4(10-25μmol ·?L-1),Ferric quinate(25 μmol ·?L-1) respectively. When both citric acid and (NH4)2SO4 were used as C and N sources,the best C N ratio was 5 : 1 or 4 : 1. As for magnetosomes forma-tion, optimal C,N and Fe sources were citric acid(50 mg per 100 mL medium)or α-ketoglu-taric acid(75-125 mg per 100 mL medium)or tartaric acid(75 mg per 100 mL medium)or sodium acetate (175 mg per 100 mL medium), NaNO3 (10 mg per 100 mL mdeium)or (NH4)2SO4(10-20 mg per 100 mL medium) ,and FeCl3(15 μmol ·?L-1) or FeSO4(15μmol· L-1),l-3% O2 concentration was required.
