黄芪多糖的分离及其免疫活性的研究

【摘要】 本文报道了黄芪多糖的分离纯化方法、理化性质、组成并进行免疫作用的研究,黄芪水提,经732(H~+)树脂初步纯化,得粗多糖,再经超滤,Sephadex G-150,透析等方法进一步纯化,得白色粉末状多糖M wt:37500。证明为α-糖甙键连接,水解多糖检出葡萄糖、半乳糖和阿拉伯糖,克分子比为:葡萄糖:半乳糖:阿拉伯糖=1:0.95:0.70。体外试验结果表明:(1)黄芪多糖可刺激小鼠脾细胞增殖,但与ConA联合刺激脾细胞时,未见有协同作用;(2)未见黄芪多糖有诱生脾细胞IL-2的作用;(3)黄芪多糖和IL-2均能增加人体LAK细胞活性,且两者合用,效果更佳,体内试验结果表明:(1)黄芪多糖可促使小鼠外周血白细胞数量增加,并且能完全纠正环磷酰胺引起的白细胞数量下降;(2)黄芪多糖对体内淋转无明显促进作用,但对CY所致免疫功能低下有一定的纠正作用;(3)黄芪多糖可增强NK细胞活性,并可完全纠正CY引起的免疫功能低下;(4)黄芪多糖可促进1g G的产生,与PWM无协同作用。

【Abstract】 This paper reports the isolation and purification of astragalan and the studies on its im-munological activities.Astragalus menbranceus(Fisch)Bge.was extracted by water and purified preliminarily by cat ion exchange resin 732(H+).The crude astragalan was gotten and then purified further by ultra-filtration Sephadex G-150 and dialysis.White powder astragalan was obtained M wt:37500.It is proved that astragalan is linked by α-glycoside bond.Quantitative analysis shows molecular ratro:glucose:galactose:arabinose=1:0.95:0.70.The tests of murine in vitro indicate that-1.Astragalan stimulates the increasing of spleen cells,but cooperation is not found when both astragalan and Con A are added.2.Astragalan does not induce spleen cells to secret IL-2.3.The activity of human’s LAK cells can be increased by either LL-2 or astragalan and cooperation is observed.The tests in vivo indicate that:1.Astraglan increases the amount of white blood cells in murine in vitro blood and redresses the discreasing caused by cyclophosphamide; 2.Astragalan can not induce in vivo lymnhocyte transformation,but can slightly relieve the immuno-logical inhibition caused by CY.3.Astragalan can increase the activity of NK cells and throughly relieve the inhibition caused by CY.4.Ig G can be induced by astragalan,cooperation is not observed when both PWM and asytragalan are added.
