Wistar鼠主动脉内皮细胞传代培养中6-酮-PGF和TXB2水平改变的年龄关系

【摘要】 正常Wistar鼠87只(雌41只,雄46只)按年龄分为4组:幼年27只(4~6周)、青年20只(16周)、中年20只(26~32周)、老年20只(56~76周)。采用贴块法对鼠主动脉内皮细胞作传代培养,于第2、4、12和20代采样共380份,分别测培养液中6-酮-前列腺素F1α(6-酮-PGF),和血栓素B2(TXB2),(T)的含量,同时还观察了四组中各代龄的细胞生长率。结果表明,生命旺盛期(幼、青年组的2、4代)细胞增长率明显高于中老年期,且P较低,T水平较高,T/P值明显增加,有利于细胞增殖,而T/P降低是衰老的表现。

【Abstract】 According to ages 87 normal wistar rats were divided into four groups: 27 rats in childhood (4-6 weeks), 20 in young (16 weeks), 20 in middle age (26-32 weeks), and 20 in old age (56-76 weeks). Using explant attachment method, endothelial cells of thoracic aortae were cultivated passing through 20 generations, the 6-keto-PGF1a and TXB2 levels in the cultivated fluids sampled from 2, 4, 12 and 20 cultivated generations were determined, and cell growth curves of all four generations of each age group mentioned above were observed. The results showed that the cell growth in 2,4 generations of childhood and young rats were far rapidly appearing as log-normal curve type, correspondly 6-keto-PGF1a level decreased relatively or absolutely and T/P ratio increased. We suggested that the increase of T/P value during the most vigorous life stage was advantageous to cell growth, while the decrease of T/P value during middle and old age was the signature of senility in vessels.
