大鼠离体卵泡释放卵子的动态过程
The dynamics of in vitro releasing process of eggs from rat follicles
【摘要】 用24孔培养板进行多个大鼠卵泡单孔培养,分别采用PMSG(孕马血清)或DES(乙烯雌酚)刺激幼大鼠卵泡成熟,同时可获得多个同步发育卵泡,分别培养在DMEM/Fl2或McCoy5A培养液中。通过单孔多卵泡培养系统,在7~14天的培养过程,能够观察到卵的体外释放动态过程。卵的释放率为35%~50%。。通过测量培养液的雌二醇和孕酮产值,证明卵泡内颗粒细胞分泌的雌二醇随培养天数的增加而升高,PMsG和DES刺激组分别从5.1增加到28.6nmol/L和从4.2增加到19.6nmol/L。但是,两种刺激卵泡成熟方案所得的孕酮的产量有所差异,在用PMSG刺激组孕酮产值随培养天数增加而减少(33.6~15.5nmol/L),而用DES刺激组孕酮产值一直在高于PMSG刺激组水平下波动。另外,本实验也证明体外培养卵泡是消耗外源性葡萄糖。卵的释放过程同时增加了组织型纤维蛋白酶原激活因子(TPA)的活性。因此,用多个大鼠卵泡单孔体外培养技术研究卵的释放动态过程以及卵的释放原理是一个简单而方便的模型。
【Abstract】 ln the present study multiple rat follicles cultured in single well of a 24-well-culture platewas studied.The ovarian follicles from immature rats were stimulated either by PMSG or by DES,in or-der to obtain follicles of similar developmental stage,which were then cultured either in DMEM / Fl2 orin McCoy’s 5A media。The reieasing process of eggs from follicles was observed through 7~14 days cul-ture。The releasing rates were 35% and 50% respectively in the DES group and PMSG group.The func-tion of rat follicular granulosa cells was studied via measurement of estradiol and progesterone in cuIturemedia。The production of estradioi increased from 5.1 to 28.6 nmol/ L in PMSG group and from 4.2 to19.6 nmol/ L in DES group during a period of l4 days culture, On the other hand,the production ofprogesterone decreased from 33.6 to l5.5 nmol/ L in PMSG group,but in DES group it remained athigher level than that in PMSG group,as the culture time increased. The in vitro culture of follicles con-sumed giucose in the culture medium,After releasing of eggs the activity of tissue-type plasminogenactivator (TPA) in culture medium increased. Therefore, The model of this in vitro multiple-follicle-cul-ture in single well of a 24-weil-cuIture plate can be used to study the dynamics of releasing process ofeggs from follicles, as well as to investigate the function ofgranulosa cells in vitro。
