大黄素对培养人系膜细胞纤维连接蛋白产生的抑制作用
Emodin inhibits the fibronectin productionof cultured kuman mesangial cells
【摘要】 本文以纤维连接蛋白(FN)为指标,观察了大黄组份之一大黄素素,对培养人系膜细胞细胞外基质产生的影响、结果用ELISA检测、免疫荧光染色以及系膜细胞小丘形成的大小、多寡等多种指标证明了大黄素能够浓度依赖性地抑制系膜细胞分泌的和细胞膜相关的FN水平,并能拮抗TGFβ对系膜细胞FN产生的刺激作用。大黄素的这一药理作用可能与大黄治疗多种慢性肾脏疾病的机理有关。
【Abstract】 Increased mesangial cell proliferationand accelerated synthesis of extracellularrnatrix are the common pathological fea-tures of various types of human and animalexperimental glomerulonephritis.Our previ-ous studies have shown that Emodin (the es-sential ingredient of Rhubarb) inhibitedmesangial cell proliferation both in vitro andvivo. In this study,the effect of Emodin onthe synthesis of fibronectin (Fn) was exam-ined. Human mesangial cells (hMC) werecultured in 10/00 FCS-DMEM supplementedwith various concentrations of Emodin fordifferent period of time (24 ̄72 h) . Cell pro-liferation was judged according to H-thymi-dine uptake. Fn secretion and deposition inhMC with or without TGFβ stimulationwere examined by ELISA and immuno-fluo-rescence techniques. The hillock formation(an in vitro sclerotic model) made by con-fluently cultured hMC was detected ,count-ed and photographed under microscopy. E-modin dose dependently inhibited hMC pro-liferation, delayed and lessened the forma-tion of hillock (Emodin 0 mg/L 172 ±17,5μg/ml 159± 10 , 10 mg/L 65 ±5. 5 , 20 mg/L 32± 3. 5 ;hillock No/well) ;The amount ofFn in the supernatants of hMC decreasedsignificantly after exposure to Emodin (E-modin 0 mg/L 2. 44± 0. 65 , 5 mg/L 2. 02±0. 31 , 10 mg/L 1. 04± 0. 06, 20 mg/L 0. 25± 0. 10). Fn deposition in Emodin-treatedcell with or without TGFβ stimulation wasalso significantly lessened by both ELISAassay and immunofluorescence staining. Inconclusion , Emodin inhibited mesangial cellproliferation and decreased extracellularmatrix production. These benificial effectsof Emodin might contribute to the therapeu-tic efficacy of Rhubarb in retarding the pro-gression of chronic renal failure.
