生物水合制取丙烯酰胺高产菌发酵产酶条件的研究
STUDY ON FERMENTATION CONDITIONS OF HIGH PRODUCING ACRYLAMIDE STRAIN USING BIOLOGICAL HYDRATION TECHNIQUE
【摘要】 本文报道了腈水合生物酶高产菌发酵产酶的适宜条件,所用菌株为R84、R310。通过正交综合设计试验确定了适合该菌株生长及产酶的培养基组分。在培养基中加入Mg2+,并以丙腈为诱导物,可明显地促进该菌产酶。R310菌株在选定的培养基及适宜的条件下,比活力平均水平达到80μmol/mg·min。用菌体直接进行丙烯腈分批加料催化水合,生成的丙烯酰胺浓度达到20%以上,丙烯腈转化率大于99%,选择性大于99%。
【Abstract】 This paper presents the appropriate fermentation condition for producing nitrile hydratase with high activity strains R84, R310. After mutual experiments,a suitable medium composition for the strain growing and producing nitrile hydratase is obtained. Additions of magnesium ions and the use of propionitrile as an inducer remarkably enhance the production of nitrile hydratase. When strain R310 is cultivated under an appropriate condition in the chosen medium, the average specific activity of nitrile hydratase comes up to 80 μmol/mg · min. Through the hydration of acrylonitrile by cell of the strain, the concentration of the acryamide thus produced is higher than 20%. Acrylonitrile conversion and acrylamide selectivity are both higher than 99%.
