胚脑组织的冷冻保存、培养和移植

【摘要】 本实验对胚脑组织进行了冷冻保存、培养和移植,以观察冻存后的胚脑组织在体外及宿主脑内继续生长、分化的情况。结果显示:培养4~5小时后细胞贴壁,1天后开始有轴突长出,在所有贴壁细胞中,有50%~58%能继续经培养后存活。把经冻存的胚脑细胞移植到幼鼠脑内,40~45天后尼氏染色切片观察,细胞形态正常,与周围宿主脑区有明显的界限。本实验证实冻存的胚脑组织复苏后仍保持继续生长、分化和建立突触联系的特性。

【Abstract】 In this experiment,cryopreservation,culture and transplantataion of fetal brain tissuewere conducted in an attempt to detect the ability of frozen neurons continuously growand differentiate.The results are as follows :Cells in culture began to attach to the dis-hes by 4-5 hours and neurite extention was seen regularly by 1 day after planting.Ofall the attached cells,50%~58% survived after planting.Frozen neural tissues were im-planted into the brain of neonatal rats and graft survival situation was observed withNissal methods,These graft cells were normal in morphology,and there was an obviousdemarcation line between these cells and host brain area.We conclude that the frozenneural cells still maintain the ability of continuing growth and differentiation.
