乙酰咪唑修饰对高密度脂蛋白结合人肝癌SMMC-7721细胞的影响
EFFECT OF ACETYLIMIDAZOL MODIFICATION ON HDL3 BINDING TO HUMAN SMMC-7721 HEPATOCARCINOMA CELLS
【摘要】 利用N-乙酰咪唑修饰高密度脂蛋白-3(HDL3)研究其对RDL3结合肝癌SMMC-7721细胞能力的影响。结果:HDL3乙酰咪唑修饰降低其竞争结合作用。这一抑制作用取决于乙酰咪唑浓度,在1mol/L时达最大抑制。结果提示HDL3中酪氨酸残基存在于HDL3结合确认部位,参与HDL3的受体结合过程。
【Abstract】 Our Previously study proved that there were high affinity HDL3 binding sites (HDL receptor) on the membrane of human SMMC-7721 hepatocarcinoma cells and binding of FITC-labelled Apo E-free HDL3 and native HDL3 binding to the cells was competitive. This study shows that treatment of HDL3 by acetylimidazol decreased its competitive binding capacity to the cells. This inhibition was dose-dependent, with maximum inhibition at 1mol/L of acetylimidazol. Our results suggest that the tyrosin residue in HDL3 is present in the HDL3 binding reeognition site and takes part in binding to its receptor.
