维甲酸诱导分化小鼠淋巴瘤SACIIB2克隆细胞的作用和机理

【摘要】 研究了RA对SACIIB2克隆细胞的促分化作用。结果表明,受RA处理后,SAC-IIB2克隆细胞形态的改变及生长抑制不因培养液中去除RA而立即消失,瘤细胞酸性非特异性酯酶(ANAE)染色阳性率明显升高,细胞内嘌呤核苷磷酸化酶(PNP)活性明显升高,而腺苷脱氨酶(ADA)活性明显降低,末端脱氧核苷酰转移酶(TdT)的表达有所减弱,而RA处理前后瘤细胞乳酸脱氢酶(LDH)同工酶谱型无明显改变,提示RA对SACIIB2克隆细胞有一定程度的诱导分化作用。对RA处理前后SACIIB2,细胞3-fos基因表达情况也作了初步观察和分析。

【Abstract】 On the basis of earlier studies on the effects of retinoie acid (RA)on differentiation of murine lymphocytic leukemia-lymphoma cell clone (SACIIB2)containingSRS-MuLV, further research has been undertaken to confirm the RA’s differentiating effect on lymphocytic leukemia cells.Studies were focused on the analysis of differentiation markers on SACIIB2 cells. It was found that the reversed morphological changes and growth inhibition of SACIIB2 cells caused by RA could be maintained after 10 days’culture in fresh media without RA.Results also showed that the percentage of positive α-naphthyl acetate esterase (ANAE) staining rate of SACIIB2 cells increased significantly. The RA-treated SACIIB2 cells exhibited a higher activity of purine nucleoside phosphorylase(PNP), a lower activity of adenosine deaminase(ADA)and a lower expression of terminal deoxynucleotidyl transferase(TdT)than thosein the control cells.The LDH isoenzyme profiles were not changed after RA treatment. The expression of c-fos product P55c-fos in the SACIIB2 cells were analysed before and after RA teratment.The results showed that the expression of P55c-fos in SACIIB2 cells was very high and was decreased a little after 2 days’RA treatment.
