CMV香蕉分离物抗血清的制备及应用研究

【摘要】 用从海南的香蕉花叶心腐病病株中获得的黄瓜花叶病毒(CMV)分离物作毒源,利用烟草进行肉毒繁殖,并通过PEG、差速离心获得了较高纯度的提纯病毒,用其对家兔间隔多次注射方法进行免疫,成功地制备出大量抗血清。琼脂免疫双扩散和间接ELISA方法测抗血清效价分别达1/128和10-6。实验结果表明,该血清和CMV有特异件,在CMV不同分离物间的反应没有明显差别。该血清被用于香蕉花叶心腐病的血清学检测研究,在SPA—ELISA实验中用做第一、第二专化抗血清的适宜工作浓度分别为10-4和10-3,在此条件下检测提纯病毒的灵敏度可达到2.4ng/mL,并通过实验方法证明香蕉汁液中近乎相同含量的病毒可被检测出来。本文还利用血清学方法对田间香蕉病株中病毒的分布,试管苗繁殖过程中病毒的传带等问题进行了研究和讨论。

【Abstract】 The CMV isolate obtained from mosaic and heart rot banana in Hainan was propagated in tabacco, and the virus was purified by cycles of differe-ntial centrifugation. The purified virus was subsequently injected into rab-bits weekly for several consecutive weeks,and a large quantity of antiserum was then obtained.Detection of the antiserum by agarose immuno double diffusion test and ELISA(indirect method)showed the titers were and 1/128 10-6,The antiserum was then used as the specific antibodies in SPA-ELISA for diagnosing of banana virus disease. Experiments indicated that this method could give such a high detection accuracy of<2.4ng/mL CMV in purified virus prepardtions that it gave the same sensitivity to detect virus in saps from banana plants, symptomless young suckers and tissue culture materials.
