尼莫地平和硫氮卓酮对大鼠脑缺血再灌流期自由基损伤的保护作用研究

【摘要】 本研究通过持续结扎大鼠一侧大脑中动脉和夹闭双侧颈总动脉60min,然后再通,制成大鼠局灶性皮质脑梗塞再灌流动物模型。在此基础上,56只大鼠随机分成7个组。观察预先用药对再灌流期脑组织脂质过氧化产物丙二醛(MDA)及超氧化物歧化酶(SOD)的影响。结果表明:再灌流后脑组织MDA含量升高,SOD活力下降,两药较小剂量组单独应用对MDA含量升高无抑制,但能抑制SOD含量降低。两药较大剂量组及联合用药组能完全抑制再灌流后脑组织MDA升高和SOD活力下降。

【Abstract】 Abstract A reperfusion model was done in rats by continuous ligature of middle cerebral artery and clipping of both common carotid arteries for 60 minutes to produce focal ischemic cerebral infarction. Fifty-six adult rats were randomly divided into seven groups. The effect of nimodipine and diltiazem on malondialdehyde(MDA)and superoxide dismutase (SOD)was studied.It was noted that the content of cerebral MDA was elevated, and that of SOD was decreased, When the drugs were used singly in smaller dosage, they didn’t exhibit inhibition on the elevation of MDA,but on decrease in SOD content.However ,when the drugs were used in combination in larger dosage, they could completely inhibit the elevation of MDA and decrease SOD activity. The authors believe that nimodipine and diltiazem may prevent free radical’s injury during reperfusion of cerebral infarction.
