B.t.CryIA基因的人工设计、改造和建构(简报)

【摘要】 为了从转录和翻译两个水平上增加B.t.CryIA基因在转基因植物中的表达,一方面利用PCgene对该基因进行了全面重新设计,删除了前面一小段和后面一大段对杀虫效果没有影响的氨基酸顺序,对剩余的约2kb的基因不改变氨基酸序列,只改变DNA序列,改造原则主要为两点:消除引起mRNA不稳定的因素和采用适宜于植物中表达的codonusage,通过对每一个氨基酸密码子进行分析,直到确定整个基因序列;另一方面我们把改造后的B.t.基因构建在两种适宜于不同植物的强化表达载体上,目前已完成设计、改造及建构工作。

【Abstract】 The B,t.CryIA gene was completely redesigned in order to enhance its expression in two lev-els of both transoription and translation in transgenic plants. Its small former and large latter regions weredeleted because of their uselessness to insecticidal activity.The rest of its amino acid sequence was retainedwhile its DNA sequence was modified according to the following two roles:the elimination of the factors of in-ducing mRNA unstability and the use of plant-prefered cedon usage. After every cedon was repeatedly anal-ysed,the gene was finally determined.In addition,the new B.t.gene was constructed in two strengthened ex-pression plasmids,which are suitable for twn different kinds of plants(monocat and dicot resperticety).Nowwe are tranfering the gene to different plants.
