芝麻体细胞胚胎发生及其内源激素和可溶性蛋白质的变化
Somatic Embryogenesis from Hypocotyl Culture of Sesame and Changes of Endogenous Hormones and Soluble Proteins During Embryogenesis
【摘要】 以芝麻(Sesamicm indicum L.)无菌苗下胚轴为外植体,在附加0.5mg·L-12,4-D和1.0mg·L-1KT的MS固体培养基上诱导出胚性愈伤组织后,转入到去掉2,4-D和KT的MS液体培养基中经悬浮震荡培养产生了胚状体,在分化培养基中诱导生根。胚状体发生于胚轴维管组织周围薄壁细胞处。在体细胞胚胎发育过程中,ZR+ZT与ABA总量比IAA高10~50倍,ZR+ZT的含量变化与ABA呈负相关,非胚性愈伤组织向胚性愈伤组织转化时IAA和ABA含量升高而ZR+ZT则相反。不同组织可溶性蛋白质分子量分别为:白色愈伤组织110kD,绿色愈伤为27和84kD,胚性愈伤组织为27和50kD,球形胚和心形胚为15、27、84kD,子叶形胚为48kD,根为55kD。
【Abstract】 Hypocotyl segments of aseptic seedlings of sesame were cultured on MS solid medium supplemented with 0. 5 mg. L-1 2,4-D and 1. 0 mg . L-1 KT. Callus was induced and then transferred into MS li-oid medium without 2,4-D and KT. Embryoids were induced by suspension culture. Embryogenesis was observed in the parenchyma around vascular tissue.During the embryoid development, the total content of ZR + ZT and that of ABA were 10 to 50 times higher than that of IAA, and the content of ZR + ZT was negatively correlative to that of ABA. The contents of IAA and ABA increased, and that of ZR+ZT decreased during the formation of embryonic callus. The molecular weights of proteins in different cultured materials are: 100 kD in whicte callus, 27 kD and 84 kD in green callus, 27 kD and 50 kD in embryonic callus, 15 kD, 27 kD and 84 kD in global and herat-shaped embryos, 48 kD in cotyledon-shaped embryos, and 55 kD in roots.
