黄瓜黑星病抗病性离体子叶接种鉴定方法

【摘要】 报道了一种新的黄瓜黑星病抗病性鉴定方法——离体子叶接种法,并对接种鉴定条件进行了研究。该方法将刚展平的子叶剪下铺到有二层滤纸的培养皿内保湿,(?)滴管将104~105个·mL-1孢子浓度的悬浮液,点滴接种到离体子叶的上表皮,每滴约0.02 mL.在15~20℃,黑暗条件下鉴定,接种后5 d 调查病情。这一方法简单、可靠、不影响幼苗继续生长。

【Abstract】 A new method for screening cucumber cultivars for resistance to scab,method of detached- cotyledon inoculation was reported.The conditions of inoculation and screening were examined.The newly-expanded cotyledons were cut and placed in petri dishes with 2 layers of fitter paper wetted with wa- ter.A 0.02 mL inoculum droplet(104~105 spores·mL-1)was inoculated on the upper surface of each cotyledon with a dropper.Inoculated cotyledons were incubated in darkened condition at(15~20)C tem- perature.The disease indexes were read 5 days after inoculation.This method was reliable and efficient and didn’t affect the growth of young plants.
