大鼠肾脏白细胞介素1受体mRNA表达的研究
Interle ukin 1 Type-1 Receptor mRNA Expression in Rat Kidneys
【摘要】 利用分子杂交原理,我们采用斑点杂交和原位杂交实验,证明了在正常大鼠肾脏有白细胞介素-1I型受体特异mRNA表达,分布于肾脏皮质和髓质。用兔抗大鼠肾皮质血清诱导产生的大鼠肾炎模型肾组织,其白细胞介素-1I型受体特异mRNA表达量明显高于正常肾组织,尤其髓质部分较为显著。肾脏固有细胞如上皮细胞及肾小球中某些细胞(可能为系膜细胞)在正常情况下有少量介素-1I型受体mRNA表达,随着炎症的产生表达量明显增高。我们的实验结果为白细胞介素-1在肾脏中的功能通过结合白细胞介素-1特异受体的作用机制,白细胞介素-1在肾脏中的自泌与旁泌作用方式,在分子水平上提供了实验依据。
【Abstract】 By using dot blot and in situ hybrluIzation, we examined interleukin 1 type-1 recepter(IL-1RtI) mRNAexpression and tissue distribution in rat kidneys of normal and nephritis。The resuits showed that fow amount of IL-IRtImRNA were expressed in normal renal cortex and medulla and incresed in the kidney of rat with nephrotoxic serumnephritis, esprially in medulla。 Some renal resident cells(epithelial cells and may be masangial cells ) were found IL-IRtImRNA cxpression and incresed in nphritis. Our data suggested that the effects of IL-1 in the kidney are mediatedthrough actions of IL-1 at spocific high affinity receptors.It provided indirect evidence IL-1 autocrine and paracrine ac-tivity。
